The present method is based on a competitive radioimmunoassay (RIA). During the incubation, the monoclonal anti-TPO antibodies in solid phase competes with the sample/calibrator anti-TPO autoantibodies for the specific sites of the 125I labelled TPO (tracer). After aspiration and washing, the radioactivity in the tubes is measured in a gamma counter. The degree of binding will be inversely proportional to the sample anti-TPO autoantibodies concentration. The calibrators of anti-TPO auto-antibodies are calibrated against the TMAb international reference preparation MRC66/387.
